Figure 2 MK is a mitogen for human endothelial and neurofibroma-derived cells. HUVEC and Hs422T neurofibroma-derived cells were plated at 1000 cells per well in a fibronectin coated 96 well plate in IMEM/10% fetal bovine serum. After 16 h, 100 μl of serum-free media, SW13, or MK6 conditioned media were added in triplicate. Cell growth was determined at day 4 after stimulation. For dose–response analysis, heparin-affinity purified MK was used (middle panel). HUVEC cells were plated in fibronectin coated 12-well plates at 5000 cells per well. After 16 h, 1 ml of serum-free medium supplemented with 50, 25, 10, and 0 μl of the 0.9 M NaCl heparin-fraction from MK6 cells and 50 μl 0.9 M NaCl fraction from control SW13 cells was added. Cell growth was determined at day 4 after stimulation.
