Screening of the Pathogen Box identified compounds with activity against human fungal pathogens. Screening of the Pathogen Box identified compounds with activity against human fungal pathogens. Each of the 400 compounds of the Pathogen Box (MMV, Switzerland) was analyzed for its impact on growth of C. neoformans wild-type strain H99 and C. albicans wild-type strain SC5314. Cultures were grown in YNB medium at 30°C with shaking for the indicated time periods, and growth was evaluated via measurements of the OD600. Results are represented as a heat map, with green indicating growth and black indicating no growth. All compounds were analyzed at a final concentration of 1 µM. The first and second rows from the tops of both heat maps represent results for the no-compound control and vehicle control, respectively. Black triangles at the sides of the heat maps indicate compounds with antifungal activity. Note that the novel compound MMV688271 completely prevented fungal proliferation. François L.
