UNISEC Correlates of Protection for Universal Influenza Vaccine Candidates Problem 1: Correlates of CMI-induced immunity unknown Solution: Measure as many as possible Problem 2: Limited number of cell aliquots ava ilable Solution: Simultaneous measurement Problem 3: Interlaboratory standardisation of cell-based assays very difficult Solution: Carry out core assay in one central laboratory CMI PBMC from vaccinated patients stimulated by antigens/peptides of choice for six hours in the presence of co-stimulatory antibodies and protein transport inhibitors. Cells stained for surface markers (CD3, CD4, CD8) Cells stained for cytoplasmic cytokines (IFNγ, IL2, IL4, TNFα) before FACS analysis.
