Figure 1 Speculation of the role of caspase-8 silencing in hepatocarcinogenesis. Death-receptor-mediated caspase-8 activation results in apoptosis and cell death. If the caspase-8 promoter is methylated inhibiting SP1 binding and transactivation then cellular fates are switched to that of tumorigenesis, potentially secondary to potentiating the Fas/NF-κB pathway (dashed line). If histone deacetylases (HDAC) or methylase inhibitors are used in this model then the fate of tumor cells can be reversed back to that of cell death.
