Comparative chromosome map of the CP-hvKP strain (NUHL30457) and 10 other hvKP strains (1158, CG43, HK787, RJF293, NTUH-K2044, RJF999, ED2, 1084, KCTC-2242 and KP52.145) using NUHL30457 as the reference. The circular map was generated using the BLAST Ring Image Generator. A sequence comparison revealed that the 11 hvKP strains were highly similar; CG43 and HK787 showed 97% coverage and 99% identity with NUHL30457. The layout of the circular diagram suggests that NUHL30457 provides a single contig representing the most completely assembled chromosomal sequence among hvKP strains. VF, virulence factors; AR, antimicrobial resistance genes. This figure appears in colour in the online version of JAC and in black and white in the print version of JAC. Whole-genome phylogeny of K. pneumoniae NUHL30457 relative to other carbapenem-resistant or hvKP strains. Scale bar indicates the distance of 0.02 nucleotide substitutions per site. The tree demonstrates that the core genome of K. pneumoniae NUHL30457 is similar to that of other K. pneumoniae isolates that have been sequenced. The table shows microbiological characteristics and virulence features of K. pneumoniae NUHL30457 and other CRKP or hvKP strains. The arrows indicate the only two CR-hvKP strains that produce carbapenemases (Kp_Goe_154414 produces OXA-48; NUHL30457 produces KPC-2 and NDM-1). Gene map of eight virulence plasmids and p30457-1 harboured by K. pneumoniae strains. Alignment of the eight virulent plasmids recovered from our strain NUHL30457, capsular serotype K2 hvKP CG43, capsular serotype K1 hvKP NTUH-K2044, capsular serotype K2 hvKP RJF293, capsular serotype K1 hvKP RJF999, capsular serotype K1 CR-hvKP Kp_Goe_154414, capsular serotype K2 hvKP KCTC-2242 and capsular serotype K2 hvKP KP52.145. Virulence factors, such as iroBCDN, iucABCD, rmpA, rmpA2 and iutA, are all located in the region corresponding to the pLVPK-like plasmid region. VF, virulence factors. Virulence characteristics of the K. pneumoniae NUHL30457 strain and the reference strains. (a) Serum resistance level of the K. pneumoniae NUHL30457 strain and the reference strains. The K. pneumoniae NUHL30457 strain and the hvKP BD2411 strain exhibited grade 6 and grade 5 responses, respectively, whereas K. pneumoniae ATCC 700603 exhibited a grade 1 response. (b) Neutrophil phagocytic rates of the K. pneumoniae NUHL30457 strain and the reference strains. Data are presented as mean phagocytic rate ± SD at 0, 5, 15 and 30 min. The phagocytic rate of the K. pneumoniae ATCC 700603 strain was significantly higher than that for either BD2411 or NUHL30457 at 5, 15 and 30 min (*P < 0.05). (c) Kaplan–Meier survival curves for K. pneumoniae NUHL30457-infected mice. Mice were intraperitoneally infected with 103 cfu of the different K. pneumoniae strains. The previously reported hvKP strain BD2411, cKP strain K. pneumoniae ATCC 700603 and saline were applied as controls. NUHL30457 showed virulence that was not significantly different from that of BD2411 (P > 0.5, by log-rank test). No mice in the ATCC 700603 or saline groups died during the 14 days.
