(c) The scheme depicts the observed interference of K16 knockout (and presumably also PC-related K16 mutation) with NRF2 activation that is overcome by SF treatment in male Krt16–/– mice, resulting in reduced palmoplantar keratoderma. These beneficial effects were not observed in female Krt16–/– mice. Simultaneous treatment of SF and diarylpropionitrile, which is an estrogen receptor-α selective agonist and estrogen receptor-β antagonist, however, led to NRF2 activation and reduced palmoplantar keratoderma.
