AG-221 can reduce intracellular 2HG levels and induce differentiation in primary human IDH2R140Q- or IDH2R172K-mutant AML patient samples treated ex vivo. Cells were cultured in the presence of AG-221 (0.1, 1, or 5 μmol/L) or DMSO (0.1%, v/v, used as a vehicle control) for up to 12 days. A, Percentage of intracellular 2HG remaining after 4, 8, and 12 days of treatment with AG-221 relative to DMSO control at the indicated doses.
