Figure 1 Angiotensin II (Ang II) induces O2• generation in intact tubular cells. Example of representative tracing of O2• measurement using the lucigenin method in LLC-PK1 (A) and MCT (B) cells. Cells were stimulated with a single dose of 10-7 m Ang II for four hours, and measurement of O2• generation was started in the luminometer after addition of lucigenin. Emission of relative light units (RLU) was corrected for unspecific luminescence in the absence of cells. Addition of the cell-permeable scavenger Tiron after five minutes of recording (↓) completely inhibited chemiluminescence in control as well as Ang II-stimulated cell. Figure 3 [3H]leucine incorporation into de novo synthesized proteins of LLC-PK1 cells as a parameter for hypertrophy. Cells were stimulated with 10-7 Ang II (▪) or treated with control medium (□) for 24hours. The flavoprotein inhibitor DIP as well as the antioxidant NAC completely abolished Ang II-mediated increase in [3H]leucine incorporation, but had no effect on basal protein synthesis in unstimulated controls. Figure 4 Western blot for p27Kip1protein. LLC-PK1 cells were stimulated for 18hours with 10-7 m Ang II in the presence of absence of 5 to 10 μm of the flavoprotein inhibitor DIP. As predicted, Ang II-treatment induced p27Kip1 protein expression. This induction was attenuated by DIP, but the flavoprotein inhibitor did not influence basal p27Kip1 expression. Figure 6 Effect of p22phox antisense oligonucleotides on Ang II-induced p27Kip1protein expression in LLC-PK1(A) and MCT (B) cells. Western blot. Quiescent cells were transiently transfected with either 1 μm p22phox sense or antisense phosphorothioate oligonucleotides using liposomes. p22phox antisense treatment almost completely abolished the Ang II-mediated stimulation of p27Kip1 expression. This effect was somewhat stronger in LLC-PK1 cells (panel A) compared with MCT cells (panel B). Interestingly, transient transfection of control cells with p22phox sense oligonucleotides induced basal p27Kip1 expression. Figure 7 Generation of O2•by xanthine/xanthine oxidase also stimulates p27Kip1expression in LLC-PK1 cells. Western blot. Quiescent cells were incubated in serum-free medium for 18hours with 5 to 10 μm xanthine or 10-7 m Ang II. The p27Kip1 protein expression induced by 10 μm xanthine was even stronger compared with 10-7 m Ang II. Co-incubation with 0.5 units/ml SOD had no effect on xanthine-mediated p27Kip1 expression. Figure 8 Proposed signal transduction pathways of how Ang II may stimulate p27Kip1expression. After binding to AT1-receptors, Ang II leads to an increase in p22phox expression. An increase in this subunit may activate enzyme activity of the membrane-bound NAD(P)H oxidase. This leads to the formation of O2•, which in turn increases the protein expression of p27Kip1. Ultimately, p27Kip1 binds to G1-phase CdK-cyclin complexes, inhibits their kinase activities, and induces G1-phase arrest leading to cellular hypertrophy. How exactly O2• stimulates p27Kip1 expression is currently unknown. Inhibition of protein degradation, stimulation of translational activity, or, alternatively, O2•-mediated TGF-β expression could all be potential mechanisms.
