Figure 1 Parameters and Issues Involved in Fragile Nucleosome Assembly at a Yeast Promoter The top part of the figure illustrates a nucleosome-free region (NFR) encompassing a promoter flanked by two stable nucleosomes or SNs. The middle shows that RSC or GRFs bind to the NFR, outwardly shifting the positions of the SNs to both create space for and to help assemble the FN. The precise composition of the FN is unknown, and several alternatives are illustrated to the right of the bracket, such as a hexasome, a heavily modified nucleosome, or an H2AZ-bearing nucleosome, although none of these is mutually exclusive. The key depicts the various components. The bottom portion illustrates a second issue: whether the FN co-exists with the Pol II machinery or whether, at any given moment, either the Pol II machinery or the FN occupies a promoter in a mutually exclusive manner.
