Volume 39, Issue 1, Pages 47-54 (July 2003) Protective effect of central thyrotropin-releasing hormone on carbon tetrachloride- induced acute hepatocellular necrosis in rats  Yoichi Sato, Masashi Yoneda, Kimihide Nakamura, Isao Makino, Akira Terano  Journal of Hepatology  Volume 39, Issue 1, Pages 47-54 (July 2003) DOI: 10.1016/S0168-8278(03)00146-6

Fig. 1 Effect of intracisternal thyrotropin-releasing hormone (TRH) analog, RX 77368, on CCl4-induced elevation of serum alanine aminotransferase (ALT) level. RX 77368 (1, 3, 5, 10 ng) was injected intracisternally 60 min before CCl4 (2 ml/kg) administration. Control animals were intracisternally injected with saline 60 min before CCl4 administration. Blood samples were collected before and 24 h after CCl4 administration. Each column represents the mean±SE of serum ALT levels. **P<0.01 compared with vehicle injection group. Journal of Hepatology 2003 39, 47-54DOI: (10.1016/S0168-8278(03)00146-6)

Fig. 2 Time course response of intracisternal RX 77368 injection against CCl4-induced liver damage. RX 77368 (1, 3, 5, 10 ng) was injected intracisternally 60 min before CCl4 (2 ml/kg) administration. Control animals were intracisternally injected with saline 60 min before CCl4 administration. **P<0.01 compared with saline injection group. Journal of Hepatology 2003 39, 47-54DOI: (10.1016/S0168-8278(03)00146-6)

Fig. 3 Effect of intracisternal RX 77368 on CCl4-induced hepatic histological changes. Saline (A) or RX77368 (5 ng) (B) was injected intracisternally 60 min before CCl4 (2 ml/kg) administration. Liver tissue was obtained 24 h after CCl4 administration and the specimens were stained with Hematoxylin–eosin (×75). CCl4 induced centrilobular necrosis surrounded by fatty degeneration (A), and this centrilobular necrosis was reduced by intracisternal RX 77368 (B). Journal of Hepatology 2003 39, 47-54DOI: (10.1016/S0168-8278(03)00146-6)

Fig. 4 Effect of hepatic branch vagotomy (A), atropine methyl nitrate (B), indomethacin (C), l-NAME (D) and 6-OHDA (E) on the intracisternal RX 77368-induced protection of the elevation of serum ALT levels by CCl4. Hepatic branch vagotomy was performed 3 days before, atropine methyl nitrate (0.15 mg/kg, i.p.) was injected 30 min before, indomethacin (5 mg/kg, s.c.) was injected 60 min before, l-NAME (10 mg/kg, i.v.) was injected 30 min before and 6-OHDA was injected 7 days before (100 mg/kg) and 4 days before (80 mg/kg), RX 77368 administration, respectively. Saline or RX 77368 (5 ng) was injected intracisternally 30 min before CCl4 (2 ml/kg) administration. Each column represents the mean±SE. *P<0.05, **P<0.01 compared with respective control group. Journal of Hepatology 2003 39, 47-54DOI: (10.1016/S0168-8278(03)00146-6)
