Figure 1 The co-incubation of resident skin cells with B. burgdorferi N40 induces an inflammatory response. (A) Dose–response of human primary keratinocytes (KCs) and fibroblasts (FBs) stimulated with various concentrations of B. burgdorferi N40 for 24hours. IL-8 (a, b), human β-defensin-2 (HBD-2) mRNA (c, d), and LL-37 mRNA (e, f) were measured as selected markers of inflammation. KCs and FBs were incubated with 100 Borrelia, 10 Borrelia, and 1 Borrelia per cell, negative control: KCs and FBs with medium alone (ø). IL-1-β was the positive control for HBD-2, and IGF-I was the positive control for LL-37. (B) Kinetic study of the inflammatory response of KCs and FBs: measure of IL-8 secretion by ELISA (g, h), measure of HBD-2 (i, j) and LL-37 mRNA expression (k, l) by quantitative real-time reverse transcriptase-PCR.
