Figure 1 Food, serum, fecal, and urinary iron levels in +/b and b/b rats. The figure summarizes iron levels in the food, blood, feces, and urine of +/b and b/b rats kept for 3 weeks on an iron-supplemented diet. This diet has previously been shown to ameliorate the severe anemia without masking the underlying defect [3]. Serum iron levels were significantly elevated in b/b compared to +/b rats (P < 0.05; N = 4). Fecal excretion and food intake were significantly higher in b/b rats compared to +/b rats (P < 0.05; N = 4). Urinary iron excretion was no different between +/b and b/b rats. Hemoglobin levels were low and no different in the three experimental groups (not shown), suggestive of no sample hemolysis. Values for ion intakes and outputs are in μmol−1/day−1· 100g body weight−1 unless otherwise stated. Figure 4 DMT1 mRNA expression inb/b rats. Medium stringency northern analysis of rat kidney total RNA (10 μg/lane) using 1.5kb DMT1 probe. Analysis was performed using 15 μg of total RNA from +/b and b/b rat kidneys (N = 4 for each group) and from the duodenum of +/b and b/b animals (positive controls). RNA was electrophoresed, transferred onto membrane, hybridized at high stringency to a 32P-labeled DNA probe common for iron response element (IRE) and non-IRE (NIRE) isoforms of rat DMT1 (nucleotides 85-1600 of rat DMT1) recognizing transcripts at 4.4, 4, and 2.4kb. Molecular weight markers (kb) are shown at the left-hand side of the figure. After detection, membranes were stripped and equivalency of loading was confirmed by using a housekeeping rat β-actin probe. IRE is iron response element. Figure 5 Semiquantitative immunoblotting DMT1 immunoreactivity in the kidneys ofb/b rats. A total of 30 μg crude membrane extracts of whole kidneys were separated by sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE) on an 8% polyacrylamide gel, blotted onto nitrocellulose membrane, and hybridized with a rabbit amino-terminus antibody against rat DMT1 sequence. The antibody was raised in rabbit against a peptide sequence (amino acid 1-21) of the putative rat DMT1, which is identical in both iron response element (IRE) and non-IRE (NIRE) isoforms, and was affinity purified as described elsewhere prior to use [14]. DMT1 immunoreactivity of crude membrane extracts from whole kidneys from +/b and b/b rats (representative of four for each group). The DMT1 antiserum recognizes a doublet of approximately 61 to 63 kD in kidney extracts from +/b animals, which is strongly reduced in kidney extracts from b/b animals.
