First breeding works at Lycium sp in UASMV Bucharest Curelaru Vlad Constantin Assoc. Prof. Dr. Adrian Asanică International Student Symposium "Hortus Academicus“ April 27, 2018 - Bucharest, Romania

Introduction The original habitat of the goji plant, often referred to as the wolfberry, is to date unknown. The region of origin is proposed to be somewhere in the south-eastern parts of Europe or the south-western region of Asia. Nowadays this plant is considered a highly valuable plant in medicine, health, treatments, horticulture production and economic investment due to the prize of the berries International Student Symposium "Hortus Academicus“ April 27, 2018 - Bucharest, Romania

A Vitamins C E Antioxidants Fiber Beta-carotene The fruit of Lycium barbarum have a variety of pharmacologic functions, life extending capabilities, anti-depressant and anti-bacterial properties.

Materials and Methods Choice of parents, buds, flowers Hybrid fruit Choice of parents, buds, flowers Flower emasculation Izolation of emasculated flowers Harvest of flower pollen Pollen conservation (if need) Breeding

Emasculation: removal of all anthers or stamens to prevent self-pollination.

Isolation of flowers: To protect them from foreign pollenm we cover the flowers with a paper bag.

Directly by touching with flowers With previous collected pollen BREEDING - crossing Directly by touching with flowers With previous collected pollen

Hybrid fruit with hybrid seeds

The seedling and emerging of hybrid seeds

Results and discussions We have 6 biotypes of Lycium sp. as parents: Matern and Patern use B1 B2 410 Only patern use AS C9 BUA International Student Symposium "Hortus Academicus“ April 27, 2018 - Bucharest, Romania

First series: extracted pollen Hybrid combination Emasculated flowers Pollinated flowers Related fruits Mature fruit % hybrid fruit ♀B1 x ♂B2 20 7 1 5 ♀B1 x ♂410 4 1. ♀B2 x ♂B1 10.08.2016 ♀B2 x ♂410 9 ♀410 x ♂B1 15 8 ♀410 x ♂B2 6,6 13 30,7 6 16,6 2. 21 24.08.2016 3 42,8 First series: extracted pollen Second series: pollination by touching directly with flowers

Series Hybrid combination Emasculated flowers Pollinated flowers Related fruits Mature fruit % hybrid fruit ♀B1 x ♂B2 20 8 6 30 ♀B1 x ♂410 16 5 1 ♀B1 x ♂Bua ♀B1 x ♂C9 2 ♀B2 x ♂B1 40 ♀B2 x ♂410 15 9 3. ♀B2 x ♂AS 10 28.09.2016 ♀B2 x ♂Bua ♀B2 x ♂C9 ♀410 x ♂B1 7 ♀410 x ♂B2 4 ♀410 x ♂AS 100 ♀410 x ♂Bua 50 It can be noticed that the best results in the third series was with 410 as maternal genitor

Poor results with extracted pollen. Series Hybrid combination Number of seeds Number of emerged plants % emerged plants ♀B1 x ♂B2 8 ♀B1 x ♂410 6 1. ♀B2 x ♂B1 25 ♀B2 x ♂410 ♀410 x ♂B1 ♀410 x ♂B2 1 66 5 7,5 32 22 68,75 2. 57 38 66,66 Poor results with extracted pollen.

Series Hybrid combination Number of seeds Number of emerged plants % emerged plants ♀B1 x ♂B2 100 9 9,0 ♀B1 x ♂410 6 ♀B1 x ♂Bua 4 ♀B1 x ♂C9 ♀B2 x ♂B1 115 1 0,9 ♀B2 x ♂410 3. ♀B2 x ♂AS 26 15,4 ♀B2 x ♂Bua ♀B2 x ♂C9 ♀410 x ♂B1 35 ♀410 x ♂B2 24 8.6 ♀410 x ♂AS 15.4 ♀410 x ♂Bua In the 3rd series of breeding only the combination with AS variety as paternal genitor had satisfactory results.

Conclusions and recommendations Very good results after using the direct flower touch pollination method. The pollen had a very high viability, around 98% to all studied biotypes. The capacity of pollen germination was between 9,43% for biotype B1, 10,45% for biotype 410 and 15,38% for biotype B2. The calendar for the start of work must be earlier, immediately after the binding for the first series of flowers, first of all due to a period of growing and fructification of hybrids by the beginning of October and also at the beginning of the fructification period (June-July) the plants present a optimal health status, the hybrid fruits growing normally.
