Background Neuropeptide Y, nociceptin , and orexin A play a role in the regulation of the hypothalamo-pituitary-adrenal (HPA) axis and food intake. Corticotrophin-releasing factor (CRF) plays a pivotal role in food intake.

The ambulatory activity of each individually housed animal was measured simultaneously using the optical beam technique. Conclusion ICV administration of NPS caused a significant increase in locomotor activity, wakefulness and reduced the amount of slow-wave sleep. iPVN injection of NPS significantly inhibited food intake 1 h after injection. This effect was short lived, with no significant differences in food intake between the groups after the first hour.

Background Orexin is able to suppress sleep and induce profound wakefulness and feeding behavior. NPS receptor is expressed within hypothalamic structures implicated in the central control of feeding behavior. Interaction between NPS and Orexin

Conclusion ICV injection of NPS increased food intake 2h after the administration. NPS neurons innervate the orexin neuronal system to increase food intake.

Conclusion The role of NPS on feeding behavior is a robust phenomenon among animal species. NPS could significantly reduce food intake during the first hour, and induce a significant rebound during the second hour. The inhibitory role of NPS on feeding appears to be independent of NPY, ghrelin and leptin pathways. But it seems to be correlated with CRF system and Orexin system.

Conclusion The increase in wakefulness during the first hour post-NPS injection was also followed by a rebound in the amount of non-REM sleep at the second hour.
