Figure 1 The effect of mTOR inhibition on two types of vasculopathy often seen in patients with chronic kidney disease. mTOR inhibitors, such as rapamycin and everolimus, stabilize atherosclerotic plaque and/or regress atherosclerosis by inhibiting VSMC proliferation, promoting protective autophagy, and attenuating nuclear factor-κB (NF-κB) activation. The protective effect of mTOR inhibitors on medial calcification was also reported. The main mechanisms include the upregulation of Klotho.
