Figure 1 Expression of CXCL2 in renal tubular epithelial cells and intravital microscopy intlr4 mutant and wild-type mice. (A) Tubular epithelial cells were isolated fromtlr4 mutant and wild-type mice as described in theMethods section. Cells were incubated for 24 hours with medium control, 1 μg/mL ultrapure lipopolysaccharide (LPS) or heat-inactivated uropathogenicEscherichia coli (UPEC). CXCL2 mRNA expression levels were determined by real-time reverse transcription-polymerase chain reaction (RT-PCR). CXCL2 mRNA levels are expressed as ratio to glyceraldehyde-3-phosphate dehydrogenase (GAPDH) mRNA expression. Results are means ± SEM from two independent experiments each performed in duplicate. (B) Quantitative analysis of intravital microscopy data show endothelial adhesion and transendothelial migration of circulating leukocytes. Values represent means ± SEM in seven mice of each group. *P < 0.05tlr4 mutant C3H/HeJ vs. wild-type mice of the C3H/HeN strain. Figure 2 Infective pyelonephritis intlr4 mutant and wild-type mice. (A) Renal bacterial load 36 hours after inocculation of uropathogenicEscherichia coli is expressed as colony-forming units (CFU) per single kidney for seven to eight mice in each group. (B) Urinary leukocyte counts were assessed by light microscopy of fresh uncentrifuged urine at 36 hours after inoculation of uropathogenicE. coli. Values represent means of 10 high power fields (hpf) per urine sample of seven to eight mice in each group. (C) Abscesses were observed on the kidney's outer surface oftlr4 wild-type but not oftlr4 mutant mice. (D) Renal histology was assessed by periodic acid-Schiff (PAS) stain, Giemsa stain for neutrophils (see insert) as indicated (original magnification 400×). (E) Total renal mRNA was extracted from six mice each of both strains and mRNA expression levels were determined by real-time reverse transcription-polymerase chain reaction (RT-PCR) as described in theMethods section. CXCL2 mRNA levels are expressed as ratio to glyceraldehyde-3-phosphate dehydrogenase (GAPDH) mRNA expression. One wild-type mouse had exceptionally high renal CXCL2 mRNA levels (arrow). Expression intlr4 mutant mice were below the detection level. Figure 3 Studies with chimeric mice. (A) Chimeric mice were generated by total body irradation and bone marrow transplantation usingtlr4 wild-type C3H/HeN mice andtlr4 mutant C3H/HeJ mice as described in theMethods section. This resulted in four mouse strains named as follows: N/N mice withtlr4 wild-type renal cells and tissues andtlr4 wild-type bone marrow-derived cells, N/J mice withtlr4 wild-type renal cells and tissues andtlr4 mutant bone marrow-derived cells, J/N mice withtlr4 mutant renal cells and tissues andtlr4 wild-type bone marrow-derived cells, J/J mice withtlr4 mutant renal cells and tissues andtlr4 mutant bone marrow-derived cells. (B) Renal bacterial load 36 hours after inoculation of uropathogenicEscherichia coli is expressed as colony-forming units (CFU) per single kidney for six to nine mice in each group. (C) Urinary leukocyte counts were assessed by light microscopy of fresh urine under a cover slip without centrifugation at 36 hours after infection with uropathogenicE. coli. Values represent means of 10 high power fields (hpf) per urine sample of six to nine mice in each group. (D) Renal microabscesses were assessed by light microscopy of GR-1-stained renal sections of mice from all groups. Values represent means ± SEM of one renal cross-section per mouse of six to nine mice in each group.
