FIGURE 1 Characterization of embryonic stem (ES)-like cells derived from the inner cell mass of tetraploid blastocysts that were aggregated with E14 diploid cells. Alkaline phosphatase (AP; A–C), anti-SSEA-1 (D–F), anti-SSEA-3 (G–I), anti-SSEA-4 (J–L), and Oct-4 (M–O) were used for the characterization. Two batches of ES-like cells (SNU-Tet1 and SNU-Tet3) were characterized, and as the positive control, a commercial ES cell line was employed. The results showed that both tetraploid-derived ES-like cells were positive for anti-SSEA-1 antibody, AP, and Oct-4, while negative for anti-SSEA-3 and anti-SSEA-4 antibodies, which was identical to the reactivity of the commercial ES cell line (scale bar, 150 μm). FIGURE 2 Microsatellite analysis of live births derived from the transfer of tetraploid blastocysts aggregated with E14 diploid cells. A total of six markers (D10Mit035, D18Mit060, D14Mite082, D14Mite096, D09Mit206, and D11Mite242) were provided, and a comparison was made among the E14 cells, surrogate mothers, live births (tail cells), SNU-tet1, and SNU-tet3. The results showed that the DNA spectra of the live births, SNU-tet1, and SNU-tet3 were identical with those of the E14 diploid cells, which differed from those of the surrogate mothers. Cho.
