1 July CoolLEDs pE LED excitation systems

1 July CoolLED CoolLED is a UK-based specialist LED technology manufacturer It is owned by Custom Interconnect Ltd. (CIL), an established electronics manufacturing company CoolLEDs Life Sciences & Analytical division supplies the pE family of LED excitation products CoolLED builds its LED products using the bare semiconductor LED die to achieve the highest possible performance. CoolLED is not restricted to commercially available packaged LED products which means it can offer a greater range of wavelengths, stability and intensity

1 July CoolLED pE -series of modular LED fluorescence excitation products with a choice from (currently) 18 wavelengths pE-1 is a 3-wavelength system pE-2 is a 4-wavelength system LEDs are carried on interchangeable LAMs (LED Array Modules) Manual Control Pod or Automation (e.g. MetaMorph) Instant On/Off and 0-100% control in 1% steps Stable, repeatable and homogeneous excitation Can be used in conjunction with other light sources Makes high speed Shutters and ND Filters redundant Product Overview

1 July CoolLED has the largest range of LED wavelengths for exciting fluorophores LED Wavelengths

1 July Largest range of LED wavelengths 2.LEDs actively-cooled for maximum intensity and stability 3.Lifetime measured in tens of thousands of on hours 4.Homogeneous excitation over FOV 5.LED Array Modules (LAMs) are interchangeable for maximum flexibility and upgrade 6.Repeatable, constant excitation during lifetime 7.No moving parts, no need to re-align 8.Fully integrated under NIS Elements + MetaMorph, MicroManager, etc. 9.Rapid Switching and Pulsing in under 300 ms 10.Cost Effective option as Shutters and ND filters redundant Key Benefits

1 July LED excitation Narrow waveband Better results from less damaged samples Less photo-bleaching Not exposing samples to unwanted wavelengths e.g. UV Stable Reliable comparisons of quantitative measurements Repeatable, reliable, and comparable results Active-cooling of LEDs using a Peltier gives best intensity and stability LED Excitation

1 July LEDs are Controllable Instant On/Off No warm up or cool down times Fast switching between wavelengths ~300 uSecs with TTL Switching does not affect LED lifetime Perfect for pre- aligning samples on a Confocal 0-100% Intensity in 1% steps Optomise intensity independently for each fluorophore Repeatable and consistent results Intensities can be pre-set for a given test LEDs are controllable

1 July Costs & Safety Long lifetime measured in tens of thousands of On hours No replacement lamps to purchase No need to re-align when replacing lamps Switching does not affect LED lifetime LEDs do not contain Mercury (Hg) which is hazardous No risk of explosion or H&S risk from Mercury vapour No Hg disposal issues. No H&S regulations to follow for operator International regulations are removing Mercury from most products Costs & Safety

1 July Better Results Quantitative Meaningful comparisons can be made between measurements made at different times Perfect for Time Lapse CoolLED excitation is stable and repeatable due to active-cooling Live Cell No vibration, no delay, no transfer of heat or electromagnetic radiation to sample Optimised intensity for each fluorophore. Dual staining applications do not require an excitation filter wheel A growing area for Fluorescence microscopy as results can be more meaningful on living cells Better Results with LEDs

1 July Mercury - Metal Halide (contains Mercury) - Xenon All suffer from the same drawbacks:- Short Lifetime Warm-up/cool-down period Decreasing Intensity Use hazardous materials H&S / Disposal issues On-going cost of lamps Lamps need re-aligned Unstable Traditional Lamps

1 July Mercury Lamps StrengthCounter Argument Broadband excitationMost needs to be filtered out More intenseOnly at some wavelengths Unwanted intensity can damage samples Mercury Lamps

1 July WeaknessAwareness Proposal Warm-up / Cool-down periodLEDs are almost instant Declining intensity through lifeLED intensity remains for tens of thousands of hours Lamps need to be replaced regularly Virtually no running costs with LED systems Lamps need re-aligned regularlyCoolLED is aligned once-ever at installation Lamps can explodeAlthough rare, it is a serious H&S risk Mercury Lamps

1 July StrengthCounter Argument Broadband excitationMost needs to be filtered out More intenseOnly at some wavelengths Unwanted intensity can damage samples Longer life than MercuryStill short by LED standards Still declines during life Metal Halide Lamps

1 July WeaknessAwareness Proposal Warm-up / Cool-down periodLEDs are almost instant Declining intensity through lifeLED intensity remains for tens of thousands of hours Lamps need to be replaced regularly Virtually no running costs with LED systems Metal Halide Lamps

1 July StrengthCounter Argument Integrated with AxiovisionAdds to investment cost as user requires latest version Simultaneous excitationOften requested but most cameras are monochrome so fast switching is actually what is required. CoolLED is developing simultaneous in the future Zeiss Colibri

1 July Zeiss Colibri WeaknessAwareness Proposal ExpensiveCoolLED pE-2 is ~6K* List with 4λ * Based on current exchange from £5375 Intensity in Green is an issuesIMP Vienna (Zeiss lab) reported that CoolLED is 4-5x intense in this region. Purchased 12 units Only integrated with AxiovisionExcludes users using NIS Elements, MetaMorph, MicroManager, etc. Latest version of Axiovision required and adds to cost Only 10 wavelengths availableCoolLED has 18 and will develop more

1 July ProductComment Thor LabsSingle wavelength Lower intensity No integration Cairn ResearchLess wavelength options Less intense FraenLess intense Transmitted light option Other LED sources All other LED product suppliers are limited to using commercially available LED packaged parts which are not actively-cooled to achieve CoolLEDs extra intensity and stability

1 July Summary Use of Mercury-based lamps will decline CoolLED LEDs offer your users many benefits including:- Instant On/Off Control of intensity from 0-100% in 1% steps No alignment or lamp replacement In many instances, there can be an overall cost saving with a CoolLED system when shutters and ND filters are removed from microscope You can contact CoolLED for advice and recommendations to ensure users get best performance (0) www.CoolLED.com
