Figure 1 Each cell of the terminator mouse contains a transgene driving expression of the human diphtheria toxin receptor (DTR) under control of the ubiquitously active ROSA26 locus. (1) To derive a specific population of target cells from a complex tissue, the terminator mouse is crossed with a Cre mouse line driving Cre recombination specifically within the target cells (2). As a consequence, the terminator transgene is inactivated within the target cells. (3) Next, the cells of the tissue are transferred into primary cell culture. (4) Unwanted cells are removed by the addition of diphtheria toxin and only the target cells survive.
