Figure 1 Detection of fibronectin and laminin in Madin-Darby canine kidney (MDCK) cells by Western blot. Cell extracts were prepared from cells cultured in collagen gel in the presence of hepatocyte growth factor (HGF) for six (lane 2) and eight (lane 3) days. Lane 1 indicates purified fibronectin (FN) and laminin (LM) isolated from human plasma and placenta, respectively, as positive controls. In MDCK cells, laminin β and γ chain and one band containing premature β and γ (pβ, pγ) chains are present. Laminin α chain is absent in these cultures. Human placenta laminin is composed of laminin α, β and γ, and merosin (M) as indicated. Figure 7 Comparison of growth rates of various fibronectin antisense transfectants cultured in collagen gel. Cells from pSG5 control (□), antisense expressing pSG5FN#1 (○), and #2 (▵) were cultured in collagen gel in the presence of HGF. At a different culture day, cells were harvested by treatment of the gel with collagenase and trypsin. Their cell number was assessed by a hemocytometer.
