Increased LIMD1 expression in hypoxia inhibits tumour growth and vascularisation Increased LIMD1 expression in hypoxia inhibits tumour growth and vascularisation AImpaired hypoxic induction of LIMD1 induction increases HRE‐luciferase activity in lung adenocarcinoma cells. Isogenic HREwt and HREmut A549 cell lines were co‐transfected with a synthetic HIF‐1‐driven luciferase (pNL‐HRE) and pGL3 firefly normalisation plasmid, prior to exposure to hypoxia. Luciferase activity was assayed and normalised against HRE activity in the HREwt line.BImpaired hypoxic induction of LIMD1 induction increases endogenous HIF‐mediated gene expression. RNA was extracted from the isogenic cell lines following 24‐h hypoxic exposure, and the expression of a panel of HIF‐1 target genes was quantified by qRT–PCR.
