Figure 1 Schematic Representation of the Mosaic-Seq Strategy (A) In a population of cells, different sgRNAs repress different enhancers by the recruitment of dCas9-KRAB, in some instances leading to a reduction of expression of a target gene. The presence of different sgRNAs is traced by a barcode that is present in the same construct as the sgRNA. (B) By combining the data from hundreds of single-cell RNA-seq (scRNA-seq) profiles for each sgRNA, the effect of each enhancer on each gene can be assessed.
