FIGURE 1 Autoradiograph of a Northern blot analysis of human epididymal RNA. Total RNA (10 μg) isolated from the testis and from the proximal caput (PCp), caput (Cp), distal caput (DCp), corpus (Co), proximal cauda (PCd), and distal cauda (DCd) regions of the human epididymis was subjected to electrophoresis through a denaturing 1.2% agarose gel, transferred, and probed with 5α-reductase type 2 and glyceraldehyde-3-phosphate dehydrogenase (G3PDH) (control) cDNAs. FIGURE 2 The coamplification of 5α-reductase type 1 (•) (A) and 5α-reductase type 2 (■) (B) mRNA with cyclophilin (▴) mRNA by PCR. Reverse-transcribed mRNA (50 ng) in the human midcaput epididymis was coamplified using a final primer concentration of 20 μM each for either 5α-reductase type 1 or type 2 with cyclophilin primers (10 μM). Products from 24 to 40 cycles were separated by polyacrylamide gel electrophoresis and stained with ethidium bromide.
