Figure 1 Effects of β-hydroxybutyrate (β-HB) (striped bars) and transforming growth factor-β1 (TGF-β1) (stippled bar) on [3H]-thymidine incorporation (A) and cell numbers (B) in human kidney 2 (HK-2) cells at 48hours.β-HB (0.1 to 10mmol/L) dose dependently decreased cellular proliferation while TGF-β1 (0.62ng/mL) also decreased cellular proliferation. The results are shown as the mean of three independent experiments performed in triplicate. Post hoc tests were performed by the Dunnett test, which compares 0mmol/L β-HB to the other groups. *P < 0.05 vs. 0mmol/L β-HB. Figure 2 Dose-dependent effect of β-hydroxybutyrate (β-HB) on collagen production in human kidney 2 (HK-2) cells at 48hours. Collagen production was assessed by [3H]-proline incorporation. Cells were transfected with Smad7 or pcDNA3Smad3ΔC as described in the Methods section. CMV-SPORT-β-galactosidase was used to control for transfection efficiency. β-HB dose dependently (0.1 to 10mmol/L) increased medium-released collagen (A) but not cell-associated collagen (B). Moreover, N-acetylcysteine (10mmol/L, fifth bar), pcDNA3Smad3ΔC (sixth bar), and Smad7 (seventh bar) reversed β-HB (1mmol/L)–induced medium-released collagen production (A). The results are shown as an average of three independent experiments performed in triplicate. Post hoc tests were performed by the Dunnett test, which compares 1mmol/L β-HB alone to the other groups. *P < 0.05 vs. 1mmol/L β-HB alone. Figure 3 Effect of β-hydroxybutyrate (β-HB) and transforming growth factor-β1 (TGF-β1) on α-smooth muscle actin (α-SMA) (upper panel) protein expression. Medium alone (lane 1), 0.62ng/mL TGF-β1 (lane 2), 10ng/mL TGF-β1 (lane 3), and 1mmol/L β-HB (lane 4) was added to human kidney 2 (HK-2) cells for 48hours. Actin (lower panel) was reprobed to control for protein loading. Total cell lysates were immunoblotted with α-SMA antibodies as described in the Methods section. It is evident that TGF-β1 (10ng/mL) increased α-SMA protein expression. However, TGF-β1 (0.62ng/mL) and β-HB (1mmol/L) did not affect α-SMA protein expression. The results are shown as an average of three independent experiments. Post hoc tests were performed by the Dunnett test, which compares lane 1 to the other lanes. *P < 0.05 versus lane 1. Figure 4 Dose-dependent effects of β-hydroxybutyrate (β-HB) on transforming growth factor-β (TGF-β) promoter (phTG5-Luc) activity and TGF-β1 mRNA expression in human kidney 2 (HK-2) cells at 24hours. For the assessment of TGF-β transcriptional activity, cells were transfected with phTG5-Luc as described in the Methods section. CMV-SPORT-β-galactosidase was used to control for transfection efficiency. TGF-β1 mRNA was quantified by real-time reverse transcription-polymerase chain reaction (RT-PCR) and expressed as the ratio between TGF-β1 and glyceraldehyde-3-phosphate dehydrogenase (GAPDH) mRNA. (A) β-HB (0.1 to 10mmol/L) dose dependently increased TGF-β transcriptional activity at 24hours. (B) β-HB (0.1 to 10mmol/L) dose dependently increased TGF-β1 mRNA at 48hours. The results are shown as an average of three independent experiments. Post hoc tests were performed by the Dunnett test, which compares 0mmol/L β-HB to the other groups. *P < 0.05 vs. 0mmol/L β-HB. **P < 0.01 vs. 0mmol/L β-HB. Figure 5 Dose-dependent effects of β-hydroxybutyrate (β-HB) on p21WAF1 (upper panel) or p27kip1 (middle panel) protein expression in human kidney 2 (HK-2) cells at 48hours. Actin (lower panel) was reprobed to control for loading. Total cell lysates were immunoblotted with p21WAF1 or p27kip1 antibodies as described in the Methods section. It is evident that p21WAF1 was dose dependently increased by β-HB (0.1 to 10mmol/L), whereas p27kip1 was dose dependently increased by β-HB (0.1 to 1mmol/L) until it reached a plateau at a β-HB concentration of 1 to 10mmol/L. Relative densities are shown as an average of three independent experiments. Post hoc tests were performed by the Dunnett test, which compares 0mmol/L β-HB alone to the other groups. *P < 0.05 vs. 0mmol/L β-HB. Figure 6 Effects of β-hydroxybutyrate (β-HB) and dominant-negative Smad3 (pcDNA3Smad3ΔC) plasmid on p21WAF1 (□) or p27kip1()promoter activity at 24hours and mRNA at 48hours in human kidney 2 (HK-2) cells. Cells were transfected with WWP-Luc, p27PF, or pcDNA3Smad3ΔC as described in the Methods section. CMV-SPORT-β-galactosidase was used to control for transfection efficiency. P21WAF1 or p27kip1 mRNA was quantified by real-time reverse transcription-polymerase chain reaction (RT-PCR) and expressed as the ratio between p21WAF1 or p27kip1 and glyceraldehyde-3-phosphate dehydrogenase (GAPDH) mRNA. (A) β-HB (1mmol/L) or dominant-negative Smad3 did not affect p21WAF1 or p27kip1 transcriptional activities. (B) β-HB (1 to 10mmol/L) did not affect p21WAF1 or p27kip1 mRNA expression. The results are shown as an average of three independent experiments. Post hoc tests were performed by the Dunnett test, which compares 0mmol/L β-HB to the other groups. Figure 7 Time-dependent effects of 1mmol/L β-hydroxybutyrate (β-HB) (upper panel) on nuclear Smad2/3 protein expression in human kidney 2 (HK-2) cells. Actin (lower panel) was reprobed to control for loading. Nuclear extracts were immunoblotted with Smad2/3 antibodies as described in the Methods section. β-HB (1mmol/L) time dependently (2 to 8hours) increased Smad2/3 protein expressions and then fell at 8 to 24hours. Moreover, N-acetylcysteine (10mmol/L) (rightmost bar) reversed β-HB (1mmol/L)–induced Smad2/3 nuclear translocation. Relative densities are shown as an average of three independent experiments. Post hoc tests were performed by the Dunnett test, which compares 0hour to the other groups. *P < 0.05 vs. 0hour; **P < 0.01 vs. 0hour.
