Figure 3 Model for p53-dependent persistent Stat3 and Jak2 activation mediating immune evasion and desmoplastic stromal response. Functional loss of p53 results in ROS accumulation, reduced Shp2 activity, and, as a result, persistent Jak2 and Stat3 phosphorylation. In addition to immune evasion, persistent Stat3 activation mediates a desmoplastic stromal response, inducing activation of the Shh pathway. Shh signals in a paracrine manner, activating PSCs and leading to collagen fiber formation and thus enhancing the desmoplastic stromal response.
