Part 1I: DNA Discovery & Replication

Discovery of DNA

Erwin Chargaff 1950s Contribution: Discovered the 1:1 ratio of base pairing (Chargaff’s Rule) G=C T=A

Rosalind Franklin 1950s Contribution: Used x- ray diffraction on DNA molecules to discover its helical shape

Maurice Wilkins 1950s Contribution: Deputy director of Franklin’s lab. Responsible for “taking” Franklin’s x- rays & showing them to Watson & Crick

Watson & Crick 1950s Contribution: Used Franklin’s x-rays to develop the 1st double- helix model of DNA

DNA Replication

Vocab Review!

Replication Watson & Crick Discovered DNA structure (kind of…not really…) Revealed how replication works Each DNA strand provides the information for complementary strand because of base pairing rules A – T G - C

When does DNA replication occur?

Where does DNA replication occur?

Cell Cycle

Replication New DNA made by the enzyme DNA polymerase During DNA replication, each strand of the original DNA is used as the template DNA polymerase synthesizes the complementary new strand DNA replication is considered semi-conservative. WHY? Keeps one original strand & makes one new strand

Activity! Draw a diagram using two different colors of DNA replication for 3 rounds.

Think! What would your diagram look like if it were conservative?

Bio Facts! The human body has about 1 trillion cells that contain the 46 chromosomes in which DNA is stored. If all of the DNA in one human cell were stretched end to end, it would form a line about 1.8 M long. The DNA that makes up a single human chromosome might be made up of more than 250 million nucleotides.
