3/5 Objective: explain the process of gel electrophoresis and the key parts that are used. DMA: Collect a worksheet “Electrophoresis Exploration Record Sheet” and a Power Supply Card. Start filling out the parts of the power supply.

STUFF TO KNOW BEFORE YOU GO! Complete reading and answer the questions for this worksheet. Go to: http://learn.genetics.utah.edu/content/labs/gel/ MAKE SURE THE SOUND IS OFF!!!! Complete the following questions about the animation: How is electrophoresis similar to thin layer chromatography? What happens to short strands of DNA in comparison to long strands of DNA? Why? What do we actually “see” after we run the electricity through the gel? LIST the five steps of electrophoresis AND summarize WHAT you need to do in each step.

Dye Lab Purpose: To practice loading materials into gel well. To better understand how electricity is used to separate molecules within substances. To learn how to interpret the outcome of the gels based on the position of the molecules within the gel. THIS ACTIVITY WILL BE DIFFERENT THAN THE ANIMATIONS: The wells will be in the middle of the gel instead of near the cathode. The substances being loaded will be indictors and dyes that have both positive and negative charges unlike DNA.

Set-Up and Run Dye Lab. Measure the pH of buffer. Load gel (samples 1-8) Record which sample you loaded in the before diagram. You will need to keep track of the left and right side of the gel. Turn on gel for 10 minutes. Measure the pH of the buffer. Make acetate drawings (mark the + and – sides) Make a color blot (mark the + and – sides)
