The difference density between this map and a Pol II crystal structure (PDB: 1WCM), both filtered to 25 Å, is in blue. A difference density calculated with a free Pol II cryo-EM reconstruction is consistent, albeit noisier (data not shown). Pol II views and regions are as previously defined (Cramer et al., 2001). The Iwr1 NLS directs Pol II import through the nuclear pore complex via Kap60/95 (NPC, 3). Assembly of Pol II with initiation factors on promoter DNA releases Iwr1 (4).
