ET-1 activates the Wnt pathway leading to osteoblast proliferation and differentiation; the calcineurin/NFATc1 pathway leading to suppression of apoptosis, which encourages cell survival; as well as other genes, that together increases osteoblast proliferation and differentiation while inhibits osteoclast activity. Fig. 2 The emerging role of ET-1 on the osteoarthritic joint. ET-1 induces osteoblastic proliferation and differentiation while inhibiting osteoclastic activity. ET-1 also increases secretion of proinflammatory cytokines and neutrophil recruitment. ET-1 acts on chondrocytes to increase MMPs production and alterations in proteoglycan production and resorption balance, leading to cartilage degradation and destruction. ET-1 also perturbs NO production in endothelial cells and VEGF secretion, which can result in endothelial dysfunction and angiogenesis.
