Figure 1 Localization of bone morphogenetic protein-6 (BMP-6) expression in human ovaries. The BMP-6 expression in normal human ovaries was investigated by immunohistochemistry. Primordial follicle (a, b), primary follicle (c), secondary follicle (d), healthy tertiary follicle (e, g,) and unhealthy tertiary follicle (f, h). (b) Negative control. Arrows indicate positive BMP-6 signal. GC = granulosa cells; TC = theca cells. Figure 2 (a–e) Effect of bone morphogenetic protein-6 (BMP-6) on inhibin α (a), inhibin/activin βA (b), inhibin/activin βB (c), FSH receptor (d), and anti-Müllerian hormone (AMH) (e) messenger RNA (mRNA) expression. The granulosa cells (GC) were cultured with BMP-6 (100 ng/mL) for 24 hours. (f) Effect of various stimuli on BMP-6 mRNA expression. The GCs were cultured with activin-A, BMP-7, or BMP-6 (100 ng/mL) for 24 hours. Total RNA was extracted from the GCs and subjected to real-time polymerase chain reaction (PCR) to determine the mRNA levels. Data were normalized to GAPDH mRNA levels. Data from three different experiments were combined and represented as the mean ± SEM relative to an adjusted value of 1.0 for the mean value of the each control. ∗P<.05 (vs. control). Bars with different letters indicate a significant difference at P<.05.
