Human dendritic cells (DCs) arise from CD34+ haematopoietic stem cells (HSCs) of the bone marrow. CD34+ HSCs differentiate into monocyte-DC progenitors (MDPs), which in turn give rise to monocytes and common DC progenitors (CDPs). CDPs differentiate into plasmacytoid DCs (pDCs) and a conventional DC precursor (pre-cDC) that produces CD1c+ DCs and CD141+ DCs132,133. In normal conditions, DC subsets patrol continuously in peripheral tissues, sense environmental factors, pick up proteins and dying cells and migrate from tissues to draining lymph nodes.
