Figure 1 Schematic diagram of probe activation. Fluorochromes are covalently coupled to a polylysine backbone. Because of the proximity of the fluorochromes, autoquenching occurs so that almost no fluorescent signal can be detected in the nonactivated state. After designated enzyme cleavage, fluorochromes are released from the carrier and become brightly fluorescent.
