Figure 1 Cultured proximal tubular cells (MKCC) were incubated for 24hours in serum-free medium without hormones or growth factors. (A) MKCC were processed as described in Methods for confocal microscopy images of indirect immunofluorescence. The biotinylated proteins labeled with streptavidin-Cy5 (1 μg/mL) strictly localized in the apical membrane (left), and actin cytoskeleton staining, labeled by using tetramethylrhodamine isothiocyanate (TRICT)-phalloidin (0.1 μg/mL), localized in both apical and basolateral membranes (right). Both confocal microscopy images represent xz sections calculated from the same set of xy images (lower band). (B) Typical immunoblot with increasing concentrations of sulfo-NHS-LC-biotin or volumes of streptavidin-agarose. Cell surface biotinylation was performed as described in Methods. Biotinylated proteins were isolated by precipitation with streptavidin-agarose and subjected to 7.5% sodium dodecyl sulphate-polyacrimide gel electrophoresis (SDS-PAGE) and immunoblotting with anti-NHE3 antiserum (serum 1568, 1:5000 dilution). (C) Immunoblots of NHE3 protein in MKCC homogenate (a) and in crude membranes of mouse kidney (b) labeled with anti-NHE3 antiserum 1568 (left), and MKCC biotinylated NHE3 protein labeled with anti-NHE3 antisera 1568 and 1566 (both, 1:5000 dilution) (right). The two immunoreactive bands of NHE3 protein have an observed mobility by SDS-PAGE of 78 and 66 kD. Figure 2 Low-dose angiotensin II (Ang II) (0.1nmol/L) increased Na+/H+ exchanger 3 (NHE3) activity via angiotensin II receptor type 1 (AT1) receptor activation in cultured proximal tubular cells (MKCC). MKCC were incubated for 24hours in serum-free medium without hormones or growth factors, and then treated for 43 minutes at 37°C with 0.1nmol/L Ang II or vehicle in the absence or presence of AT1 receptor inhibitor added 5 minutes before [AT1inh = losartan (10 μmol/L) or candesartan (1nmol/L)]. NHE3 activity was measured by H+-driven initial rate of 22Na. NHE3 activity was defined as the activity resistant to 50 μmol/L Na+/H+ exchange inhibitor cariporide (HOE642) and sensitive to 300 μmol/L ethyl isopropyl amiloride (EIPA). Values are mean ± SE of 9 experiments. *P < 0.001 by paired t test. (□, control; ▪, Ang II). Figure 3 Low-dose angiotensin II (Ang II) increased apical Na+/H+ exchanger 3 (NHE3) protein abundance via angiotensin II receptor type 1 (AT1) activation but did not affect apical sodium glucose cotransporter 1 (SGLT1) protein abundance in cultured proximal tubule cells (MKCC). (A) Typical blot of biotinylated NHE3 protein (left), NHE3 protein in homogenate (middle), and biotinylated SGLT1 protein (right). (B) Summary of results: MKCC were incubated for 24hours in serum-free medium without hormones or growth factors. Then, cells were pretreated with 0.1nmol/L Ang II or vehicle for 43 minutes in the absence or presence of losartan (10 μmol/L added 5 minutes before). Cell surface biotinylation was performed with sulfosuccinimidyl-6-(biotinamido) hexanoate (sulfo-NHS-LC-biotin) (1mg/mL) as described in Methods. Biotinylated proteins were isolated by precipitation with streptavidin-agarose and subjected to 7.5% sodium dodecyl sulphate-polyacrimide gel electrophoresis (SDS-PAGE) and immunoblotting with anti-NHE3 antiserum (serum 1568, 1:5000 dilution) or rabbit polyclonal anti-SGLT1 antiserum (1:50,000 dilution). Results are mean ± SE of 3 experiments. *P < 0.05 by paired t test. (□, control; ▪, Ang II). Figure 4 Low-dose angiotensin II (Ang II)–induced increase in apical Na+/H+ exchanger 3 (NHE3) protein abundance was not due to decrease in NHE3 internalization. (A) Typical blots. (B) Summary of results: cultured proximal tubule cells (MKCC) were incubated for 24hours in serum-free medium without hormones or growth factors. Then, cells were incubated for 1hour with sulfosuccinimidyl 2-(biotinamido)-ethyl-1, 3-dithiopropionate (sulfo-NHS-SS-biotin) (1mg/mL), treated at 37°C with 0.1nmol/L Ang II or vehicle for 43 minutes, or maintained at 4°C, and incubated with 2-mercaptoethanesulphonic acid (MESNA) as described in Methods. Results were compared with cells not treated with MESNA and expressed as internalized NHE3 protein in percent of apical NHE3 protein. Results are mean ± SE of 4 experiments. *P < 0.01 vs. control by paired t test. (□, control; ▪, Ang II). Figure 5 Low-dose angiotensin II (Ang II) increased apical Na+/H+ exchanger 3 (NHE3) protein abundance by exocytic insertion activation. (A and C) Typical blots. (B and D) Summary of results: cultured proximal tubule cells (MKCC) were incubated for 24hours in serum-free medium without hormones or growth factors. Then, cells were incubated for 1hour with sulfo-NHS-acetate (1.5mg/mL) and immediately incubated with sulfosuccinimidyl-6-(biotinamido) hexanoate (sulfo-NHS-LC-biotin) at 4°C (A and B) or treated with 0.1nmol/L Ang II or vehicle for 43 minutes (C and D). Cell surface biotinylation was next performed with sulfo-NHS-LC-biotin (1mg/mL) as described in Methods. Results are mean ± SE of 3 experiments and expressed in percent of control. *P < 0.01 and **P < 0.05 by paired t test. (□, control; ▪, Ang II).
