Plk1 inhibition affects the NuMA turnover at the spindle pole. Plk1 inhibition affects the NuMA turnover at the spindle pole. (A–D) Untreated HeLa cells (control) (A, C) or cells treated with Plk1 inhibitor BI 2536 for 20 min 300 nM (B, D), fixed, and stained for NuMA (grey), and used either for spindle pole (sp) intensity measurements or cytoplasmic (cyto.) intensity measurements. DNA is shown in blue. Note that while capturing NuMA images for spindle pole (sp) intensity measurements, the spindle pole signal was not saturated in comparison with either cortical (as shown for Fig 1) or cytoplasmic NuMA quantification as shown for (C) and (D). More than 50 cells were analyzed in each condition and the represented cells are shown here. Quantification of the spindle pole enrichment was performed area of size 1.6 μm2 (shown in yellow) and quantification of cytoplasmic intensity was calculated for a rectangular area equivalent of 1.8 × 4 μm adjusted for the background signal from 10 representative cells (P < 0.0005 between control and BI 2536–treated metaphase cells for the spindle pole and P < 0.005 for the cytoplasmic levels in controls and BI 2536–treated cells; error bars: SD). (E–H) FRAP analysis of HeLa cells stably expressing mCherry-H2B at metaphase and transfected with GFP-NuMA (E, G) and also treated with BI 2536 (F, H). GFP signal is shown in grey using inverted confocal images. The unbleached and bleached region of the cell cortex is shown by the orange and blue circles, respectively. Recovery profile of the bleached spindle-pole signal is plotted for 260 s for all conditions. (t1/2) for GFP-NuMA = 27.1 and (t1/2) for GFP-NuMA + BI 2536 = 52.7 s. Please note the significantly slow recovery of GFP-NuMA at the spindle pole in cells treated with Plk1 inhibitor BI 2536. P < 0.005 between untreated control and BI 2536–treated GFP-NuMA–expressing cells at the spindle poles as calculated by two-tailed t test. (I) Immunoprecipitation (IP) using GFP-Trap from Nocodazole-arrested mitotic HeLa cells extracts expressing GFP-NuMA1,411–2,115 in the absence and presence of Plk1 inhibitor BI 2536. The resulting blots were probed for GFP and LGN as indicated. IN: 3% of total, IP: 33% of the total. Note that GFP-NuMA1,411–2,115 interacts with LGN with the similar strength in the absence or presence of Plk1 inhibitor. (J) GST pull-down assay using recombinantly purified GST-tagged Plk1-PBD using Nocodazole-synchronized mitotic cells in the presence and absence of LGN. Asterisk on the Ponceau stained nitrocellulose membrane depicts GST-tagged PBD of Plk1. Note Plk1-PBD pull-down endogenous NuMA even when LGN is absent in the mitotic lysates as detected by Western blotting using NuMA-specific antibodies.
