FACT does not disassemble nucleosomes assembled on α-satellite DNA. FACT does not disassemble nucleosomes assembled on α-satellite DNA. Nucleosomes were reconstituted with 147-bp α-satellite DNA and X. laevis histones. Nucleosome concentration was kept at 20 nM. FACT was added at the indicated concentrations, and samples were incubated at 40°C for 1 h. Samples were analyzed by 5% PAGE and visualized by SYBR Gold. In the absence of FACT, nucleosomes remain intact at 40°C (lane 2). As FACT is titrated, the amount of nucleosome and free DNA is unchanged (lanes 3–8). Tao Wang et al.
