Knockdown of Meis1 reduces the expression of Foxn4 and Lim1+2.Chick retinas were electroporated with shMeis1-1 or shControl construct at E4. Transfected retina tissues were harvested at E7, sectioned, and immunostained for cell specific antibodies: Foxn4 and Lim1+2 (green). RFP+ cells show a dramatic reduction of Foxn4 and Lim1+2 expression in the shMeis1-1 (B,D) transfected cells, but not in the shControl transfected cells (A,C). Double labeled cells (Foxn4+/RFP+ or Lim1+2+/RFP+) were indicated by arrowheads, while arrows represent RFP+ cells that were negative with Foxn4 or Lim1+2 staining. A histogram (E) shows that there was a dramatic reduction of the percentage of RFP+ cells with Foxn4 and Lim 1+2 staining in shMeis1-1 transfected population.
