(A) The C-terminal tail of the P2Y1 receptor was either deleted (P2Y1-334Z) or used to replace the C-tails of the P2Y2 (P2Y2/P2Y1 CT) and BK2 (BK2/P2Y1 CT) receptors, and the altered receptors were stably expressed in MDCK(II) cells. Localization of the truncated and chimeric receptors was analyzed in polarized monolayers by confocal microscopy. In these and subsequent images, the larger image in each panel was taken in the X-Y plane parallel to the monolayer, whereas the smaller image below the X-Y micrograph is an X-Z stack that shows a cross-section of the cells along the narrow white line shown in the X-Y image. Green fluorescence corresponds to the HA-tagged receptor, and red fluorescence corresponds to ZO-1 at the tight junction.
