The degradation of SMN proteins meditated by cysteine proteases was partially inhibited by Z-FA-FMK and E64d. The degradation of SMN proteins meditated by cysteine proteases was partially inhibited by Z-FA-FMK and E64d. 20 μM Z-FA-FMK or E64d were delivered to cell cultures after overexpression of combined proteases (CAPN1 + CAPN7, CTSB + CTSL, and CAPN1 + CAPN7 + CTSB + CTSL) for 24 h. The cells were also treated with DMSO (vehicle control) for 24 h and collected for Western blot analysis. (A–D) Western blot results from triplicate samples and quantification data showed that overexpression of cysteine proteases significantly degraded both Myc-SMN2a (A, B) and Myc-SMN2d (C, D) proteins, which was partially mitigated by the treatment of Z-FA-FMK and E64d.
