Fig. 2 Noncompetitive RT-PCR for IL-8 gene expression (A) and for the housekeeping gene hypoxanthine guanine phosphoribosyl transferase (HPRT) (B). Lane 1 contains a 100 bp DNA molecular weight ladder. Lanes 2 through 5 represent RT-PCR data with input RNA from 4 different samples of normal sinus mucosa. Lanes 6 through 14 represent RT-PCR data with input RNA from samples of sinus mucosa from 9 different subjects with chronic sinusitis. Fig. 3 Symptom scores and sinus CT scores from patients with sinusitis who have positive mucosal IL-8 gene expression are significantly higher than those from patients with no detectable IL-8 gene expression. Heavy horizontal lines represent median values. Lower and upper limits of the boxes show 25th and 75th percentile values, and lower and upper bars show ranges. Fig. 4 Representative example of data from a competitive RT-PCR for IL-8 gene expression. Lane 1 contains a 100 bp DNA molecular weight ladder. Lanes 2 through 6 show results obtained by using serial 3-fold dilutions (from 300 pg to 3 pg) of truncated IS cRNA as input. Lanes 7 through 11 are results obtained when a fixed level of test input RNA from a sample of sinus mucosa is incubated with the same range of dilutions of IS cRNA. Lane 12 shows the results when the same amount of test input RNA was subjected to RT-PCR in the absence of any internal standard. Product from authentic IL-8 mRNA is seen at 284 bp, whereas product from the IS cRNA is seen as a band at 189 bp. Competition is observed between the test RNA and the IS. In this instance equivalence was calculated to occur at an IS cRNA level of 22 pg (between the 30 pg and 10 pg levels of IS in lanes 9 and 10 ).
