Human embryonic pancreas specification. Human embryonic pancreas specification. (A) Carnegie stage 10. The schematic (sagittal) shows the proximity between the dorsal wall of the foregut endoderm (fe; blue) and the notochord (noto; red) at the level of the anterior intestinal portal (AIP; dashed line), which allows patterning of that region of dorsal foregut endoderm to a future pancreatic fate. The bright-field immunohistochemistry images show transverse sections at this level of the AIP for PDX1, SHH and FOXA2. FOXA2 marks the foregut endoderm and anterior neural tube (nt). PDX1, the key pancreatic transcription factor, is not yet expressed; however, the asterisked area in the PDX1 panel shows the region of presumptive pancreatic endoderm where SHH is already excluded. (B) Early Carnegie stage 13. The schematic (sagittal) shows the first signs of budding of the dorsal pancreas (dp; yellow) from the foregut endoderm (fe)/early embryonic duodenum (duo). By this stage, the liver bud has already extended ventrally as hepatic cords (hc; pink), the extrahepatic biliary duct (ehb; green) and gall bladder (gb; green). The immunohistochemistry sections (sagittal) show staining for FOXA2 (marks all of the structures), SOX9 (most clearly marks the foregut/duodenum, gallbladder and part of the extrahepatic biliary duct) and NKX6.1 (marks endocrine competent endoderm of the dorsal pancreatic bud). The lack of clearly visualized ventral pancreatic bud (vp; yellow) in the immunohistochemistry sections is due to the plane of tissue sectioning. Counterstaining is with Toluidine Blue.
