FIG 3 Effect of azelastine on the cytosolic calcium response or on histamine release from basophils or mast cells stimulated with an optimal concentration of anti-IgE antibody. Cells were labeled with fura-2 and analyzed for free cytosolic calcium by digital microscopy. The average net calcium elevation for the first 15 minutes of the reaction was determined from a field of 30 to 100 cells. For cells treated without azelastine, dimethyl sulfoxide at a relevant concentration was used as a control. Histamine release was determined from the supernatants obtained from the microscope chamber from which the calcium measurements were made.
