Figure 1 Western blot for the detection of the insulin receptor (IR) by colorimetry, and IR tyrosine autophosphorylation by chemiluminescence, in the ovarian tissue of a woman with polycystic ovary syndrome (PCOS) and a control woman. The left panel indicates the presence of total and β-subunit IR. Lane 1 shows the rainbow marker; lane 2 the biotinylated standards; lanes 3, 4, 5, and 6 the samples of total protein fraction from the ovaries; and lanes 7, 8, 9, and 10 the same samples after purification for IR. Bands at approximately 95 kd and 230 kd correspond to the molecular weights of the IR β-subunit and total IR, respectively. Lanes 3, 4, 7, and 8 have one-half the protein concentration of lanes 5, 6, 9, and 10. The right panel indicates the results of the Western blots using chemiluminescence to determine the degree of tyrosine autophosphorylation of the IR. Lane 1 shows the rainbow marker; lane 2 the biotinylated standards; lanes 3, 4, 5, and 6 the samples of total protein fraction from ovary; and lanes 7, 8, 9, and 10 the same samples after purification for IR. Lanes 4, 6, 8, and 10 contain the samples prior to insulin stimulation, and lanes 3, 5, 7, and 9 the samples after insulin stimulation. As expected, the insulin-stimulated samples (lanes 3, 5, 7, and 9) have darker 95-kd bands than the non-insulin-stimulated ones (lanes 4, 6, 8, and 10), which is consistent with an increase in the amount of tyrosine phosphorylation (i.e., autophosphorylation) of the IR by insulin binding.
