Simultaneous mutation scans for MYD88 and CCND3 hotspot mutations are possible using ddPCR. Here we show the results derived from the application of two probes targeting MYD88 L265P (HEX) and CCND3 (FAM) hotspot mutations across a cohort of 15 fresh tumor lymphoma samples with previously known mutations. Negative samples exhibit only three fluorescence clusters: one green cluster for MYD88 wild-type alleles, a blue cluster for wild-type CCND3 alleles and a third cluster just reflecting empty droplets (in black). Samples positive for either MYD88 or CCND3 exhibit an additional cluster showing increased fluorescence either in the FAM or the HEX channel, respectively.
