(A) Effects of INTS11 knockdown on Cajal body components. INTS11-depleted HeLa cells were fixed with 4% PFA, and coilin (green) and SMN (red) were immunostained using their specific antibodies. (B) Percentage of cells with nucleolar accumulation of coilin and cytoplasmic granules containing SMN in control RNAi-treated cells and INTS4 RNAi-treated cells. The values are means ± s.d. (error bars) of five independent experiments. More than 200 cells were counted in each experiment. (C) RNA immuno-FISH of U2 snRNA (green) was performed on INTS11-depleted HeLa cells. Arrow indicates intense U2 snRNA signals in Cajal bodies. (D) The location of the RNA probe for premature U2 snRNA used in the RNA protection assay (RPA) is shown.
