Volume 125, Issue 2, Pages 633-634 (August 2003) Differential p38 mitogen-activated protein kinase target phosphorylation in responders and nonresponders to infliximab  Georg H Waetzig, Philip Rosenstiel, Susanna Nikolaus, Dirk Seegert, Stefan Schreiber  Gastroenterology  Volume 125, Issue 2, Pages 633-634 (August 2003) DOI: 10.1016/S0016-5085(03)00979-X

Figure 1 (A) Levels of (phosphorylated) p38 MAPK, ATF-2, and Hsp27 in the colonic mucosa of infliximab-treated CD patients. Representative results obtained from patient 2 (nonresponder, NR), patient 3 (short-responder, SR), and patient 5 (responder, R) are shown. (B) Identification of apoptotic lamina propria cells in CD patients treated with a single infusion of infliximab. Before infusion (b), practically no apoptotic cells were detected, while many apoptotic (cleaved caspase-3-positive) macrophages (M) or neutrophils (N) were visible 24 hours after infusion (24) in both responders and nonresponders. Immunofluorescence staining was performed as described previously.2 Filled arrowheads, cells positive for cleaved caspase-3 (anti-cleaved caspase-3, 1/100, Cell Signaling Technology); open arrowheads, cell markers (M-b, M-24: anti-CD68/Ki-M6 for macrophages, 1/500, Dianova; N-b, N-24: anti-pan-human neutrophilic peptide for neutrophils, 1/1,000, Novocastra); arrows, colocalization of cleaved caspase-3 and cell markers; bars, 50 μm.
