Diagrammatic representation of the LMPCR footprinting technique. Diagrammatic representation of the LMPCR footprinting technique. After methylation by DMS, genomic DNA is cleaved with piperidine. Three nested oligonucleotides (primers) are then used in the LMPCR reaction to quantitate the abundance of individual piperidine-cleaved fragments (24,35). Primer 1 is used in the first strand synthesis reaction, which is followed by blunt-end ligation of a double-stranded linker.
