Interacting effects of 22(R)-hydroxycholesterol and 9-cis-RA with fatty acids and ketone bodies on the expression of ABCA1 in HepG2 cells and RAW264.7 cells. Confluent hepatocytes (A and C) and RAW264.7 cells (B and D) were incubated for 18 h with serum-free medium in the presence of 10 g/l BSA and 10 μmol/l 22R-HC (A and B) or 10 μmol/l 9-cis-RA (C and D) plus 100 μmol/l EPA, 500 μmol/l palmitic acid (PA), 500 μmol/l oleic acid (OA), 10 mmol/l hydroxybutyrate (HB), or 10 mmol/l acetoacetate (AA). The mRNA of ABCA1 was demonstrated by quantitative real-time PCR. Note that EPA, oleic acid, and acetoacetate (more prominent in macrophages, but not palmitic acid and hydroxybutyrate) (B) suppress the stimulatory effect of 22(R)-HC on ABCA1 gene expression. EPA and oleic acid have less suppression on ABCA1 expression in the presence of 9-cis-RA than in 22(R)-HC.
