Figure 1. Basal cysteine S-sulfenylation is increased in OA chondrocytes. Confluent cultures of articular chondrocytes were changed to serum-free media and the next day lysed with or without dimedone in the cell lysis buffer as detailed in the methods. Samples of cell lysates with equal amounts of total protein were separated by SDS-PAGE and immunoblotted with an antibody against dimedone-conjugated S-sulfenylated cysteines. Figure 2. FN-f stimulates Src activity and Src activity is required for FN-f induction of MMP-13. A Articular chondrocytes were transfected with a FRET-based Src activity biosensor and Src activity was measured as described in the methods. Briefly, fluorescence photomicrographs of live cells were acquired for 10 minutes prior to treatment, then for an additional 35 minutes following treatment with equal volumes of PBS, FN-f (1 μM final concentration) or EGF (50 ng/mL final concentration). Data presented represents mean response of 57, 61, 69, 42 and 56 individual cells from a single donor for PBS untreated control, FN-f, EGF positive control, FN-f +SU6656 and FN-f RV mutant negative controls, respectively. B Articular chondrocytes were pre-treated for 30 minutes with the indicated concentration of one of three different Src inhibitors (SU6656, Src Kinase Inhibitor 1, or PP2) or varying concentrations of Dimedone. Following overnight treatment with 1 μM FN-f or 25 μg/mL IL-1β, samples of equal volumes of conditioned media were separated by SDS-PAGE and immunoblotted with an antibody to MMP-13.
