Wild-type and db/db mice were treated with berberine, and basal blood glucose (A) and glucose tolerance (B) were measured. db/db mice spontaneously develop basal hyperglycemia as shown in the control (db/db-Con) versus wild-type groups. A: Basal hyperglycemia was significantly reduced in db/db mice gavaged with berberine (BBR; 560 mg · kg−1 · day−1) for 7 days, compared with controls (vehicle-only gavage). B: Glucose tolerance test. Intraperitoneal bolus glucose (1 g/kg) was given to wild-type (Wt) mice, and glucose loads to db/db mice were then matched to the lean wild-type littermates. C–F: Berberine was also administered to chow-fed and high-fat–fed rats followed by measurement of body weight gain (C), food intake (D), basal plasma triglyceride (E), and whole-body insulin sensitivity as determined by hyperinsulinemic euglycemic clamp (F). Rats were gavaged with vehicle (Veh) or berberine (380 mg · kg−1 · day−1) for 2 weeks before (but not including) the acute clamp study. For clamp studies (insulin infusion 0.25 units · kg−1 · h−1), jugular and carotid cannulae were implanted 7 days previously and animals were studied over 2 h in the conscious state after 5–7 h fasting.
