Supplementary File A Table S1: 20 live Demodex mites can modulate SZ95 sebocytes on a gene level after 8hr challenge. RT2 profiler TLR PCR arrays were utilised for gene expression analysis. This contained 84 primer sets specific for genes involved in TLR signalling events. ΔΔCt based fold-change calculations were performed for normalised mite treated cells (20 live mites) compared to untreated controls at each time point using SABiosciences web based software package (http://www.sabiosciences.com/pcrarraydataanalysis.php). Results are represented as fold change. A trend of down regulation in many genes in the TLR pathway was evident after 8hr challenge with Demodex with NFKB1 significantly down regulated by Demodex (p=0.022). Student t-test was performed for statistical significance.

B Table S2: 20 live Demodex mites can modulate SZ95 sebocytes on a gene level after 24hr challenge. Many genes in the pathway remained down regulated at 24 hr challenge with 20 live Demodex. CD180 was significantly down regulated (p=0.0489). Student t-test was performed for statistical significance.

C Table S3: 20 live Demodex mites can modulate SZ95 sebocytes on a gene level after 48 hr challenge. A trend to positive regulation of genes in TLR signalling was appearing at 48 hr, with a significant increase in CCL2 (p=0.0479). Student t-test was performed for statistical significance.
